How to make Rhizobium culture?

Rhizobium culture:

  1. Nitrogen fixation is carried by Rhizobium.
  2. Culture procedures for Rhizobium are given below:
    1. Gently fill three sterile test tubes with the mannitol-agar medium.
    2. Use sterile droppers to place a few drops of the sample (rhizobium water) into the test tubes.
    3. Mix the ingredients well.
    4. Fill various Petri plates with the mixture, then incubate at 45 degrees Celsius.
    5. When the medium solidifies, rotate the petri dish.
    6. Give the culture roughly three days to incubate.
