Solution

The correct option is B Selection and isolation of somatic cells
Protoplast fusion is a technique to get hybrid plants with desired characters. The protoplast obtained from the parent plants are fused together to form a hybrid cell which is cultured on nutrient media. The steps of protoplast fusion are as followed:
  1. Isolation of protoplast - The protoplast is isolated by breaking the cell wall using lytic enzyme on the cell wall.
  2. Fusion of protoplast - The protoplast obtained from the parent plants are fused together to form a hybrid cell which is cultured on nutrient media.
  3. Culture of hybrid protoplast on nutrient media - The hybrid protoplast is cultured on the nutrient media.
  4. Formation of callus - The hybrid protoplast undergoes cell division and forms callus.
  5. Morphogenesis - The callus is transferred to a medium containing auxin and cytokinin. Auxin and cytokinin are used in equal quantity in the growth media of callus it promotes shoot and root formation and finally leads to organogenesis.
So, the correct answer is option B.
