The graph plotted between the rate of photosynthesis (amount of oxygen evolved) against wavelength of light is called the action spectrum. Engelmann discovered the first action spectrum of photosynthesis by using Cladophora algae. He concluded that the rate of photosynthesis was maximum in red and blue light as more oxygen was released by algae which absorbed blue and red light.

Van Niel conducted experiments in purple green bacteria and demonstrated that the oxygen evolved during photosynthesis comes from water but not carbon dioxide.
