The chloroplast is double membrane covered cell organelles. They can be isolated in intact form through the use of differential centrifugation by subjecting the homogenate to successively higher centrifugal force. Most commonly used solution is sucrose or percoll density gradient column for suspension. Post nuclear fraction is called the Mitochondrial fraction. It is obtained when post-nuclear supernatant is subjected to the centrifugal force of 12000 x g for 20 minutes. This fraction comprises of mitochondria, chloroplasts, lysosomes and microbodies. Special attention needs to be devoted to avoiding osmotic shock which can lyse cell organelles. Thus, the correct answer is option D.
