DNA libraries:

  1. A DNA library is a collection of clones that contains all the fragments obtained from any organism.
  2. It can be used for constructing vectors, genetically modified organisms, or simply for screeing for a particular gene.

Screening a Library: It is the process of identifying the clones carrying the gene of interest.

Two parts of screening a Library:

  1. For screening a DNA library, the DNA from the desired organism is isolated, fragments by gel electrophoresis, inserted into vectors making clones of the fragment of DNA in bacterial cells, and then identify the sequence of DNA fragments and store this sequence computer using bioinformatics.
  2. A DNA library can be screened by denaturing a labeled probe, denaturing the vector, and inserting it into single-stranded DNA which should be done at a specific temperature at which the probe hybridizes with the target DNA.
  3. The lower the temperature, the less will the hybridized sequence obtained, and the higher the temperature, the more will be the hybridized sequence obtained.
