Hot start PCR:

  1. Hot start PCR is a technique that begins the polymerase reaction at or above the annealing temperature of the primer.
  2. This eliminates the possibility of non-specific annealling of primer extensions at ambient temperature where the Taq Pol retains partial activity.
  3. High yield and specificity of the intended gene product can be achieved with hot start PCR.
  4. The polymerase should be added when the mixture reaches a temperature higher than the annealing temperature to achieve the hot start PCR condition.
