Restriction enzymes are molecular scissors which cleave the DNA molecules at specific points.

Nucleotides are the monomeric unit of DNA. A nucleotide of DNA comprises of a deoxyribose sugar, a nitrogenous base and phosphate group. Two nucleotides are connected with each other with the help of phosphodiester bonds.

Phosphodiester bond is formed between the OH group at the 3' carbon of the deoxyribose sugar of first nucleotide and the phosphate group which is attached to the 5' carbon of the deoxyribose sugar of the second nucleotide.

Restriction endonucleases cleave these phosphodiester bonds between the two adjacent nucleotides of the DNA molecule in both the strands within or near the specific sequence.
