Whenever there is a pathogen infiltration or they are able to surpass first and second line of defence, the third line of defence comes (immune cells like macrophages, lymphocytes) comes into play.

Helper T cells get activated and secrete cytokines and in turn activate naive B cells which produces plasma B cells and memory B cells which are specific to the antigen.

Plasma B cells live for a few days and secrete enormous amount of antibodies during this time.These antibodies travel in the blood and mark the antigens to be engulfed by macrophages.

B cells mediated immunity is called humoral immunity.

Cell mediated response includes cytotoxic T cells, helper T cells and memory T cells.

Helper T cells are the most numerous T cells which does not kill the cells but activates B cells and cytotoxic T cells to kill pathogens or infected cells.

Memory T cells are generated in response to pathogen attack during first exposure to avoid or prevent the infection on subsequent exposure.

Cytotoxic T cells play an important role in organ rejection. They are killer cells. They recognise the MHC complex (protein receptor) found on every cell and this help to differentiate the foreign tissues from self, thus attacking and killing the non self.
