According to assertion statement fine structures can be observed by Transmission electron microscope (TEM).

The reason statement, study of live cells is not possible through TEM. The reason being is that the specimen will be under high pressure in Electron microscope chamber. And also sample is prepared by covalent crosslinking of gluteraldehyde and osimum tetraoxide later by dehydrating and embedding in plastic.

Only the fixed specimen can be visualized clearly using TEM
