Immobilisation of enzymes is the process of fixing enzyme to or enclosing it in a solid support so as to use the enzyme in a continuous process. When the enzyme is immobilised using the method of covalent bonding, the enzyme gets inactivated. The amino acids in the enzyme are involved in the bond formation. This results in the change in the conformation of the enzyme. The enzyme to be active needs to be present in the functional conformation which is disturbed due to covalent bonding.
