According to the chemiosmotic hypothesis, ATP synthesis is linked to the development of a proton gradient across the membrane of the thylakoid of the chloroplast. There are several processes that take place during activation of electrons and their transport which lead to the development of a proton gradient. Photolysis of water is the first step of ATP synthesis. The splitting of water molecule takes place on the inner side of the membrane and so the hydrogen ions (protons) that are produced, they accumulate within the lumen of the thylakoids.
Hence, both Assertion and Reason are correct and Reason is the correct explanation for Assertion.
