Role of cyanogen bromide
Cyanogen bromide is an inorganic compound which is used during the production of recombinant insulin, humulin.

Humulin production involved the separate insertion of the chemically synthesised genes encoding human A and B peptides of insulin into a plasmid vector for expression in the host bacterium, E. coli. These genes are inserted adjacent to the gene encoding β-galactosidase in the plasmid. Separate batches of E. coli cells produced proinsulin chains, i.e., fused β-galactosidase-A chain and β-galactosidase-B-chain. These chains were separated from the stretch of β-galactosidase by the treatment of cyanogen bromide (CNBr).

Addition of an extra codon for methionine at N-terminal of each gene for the A and B chain helped in the action of CNBr and separation.

The A and B chains after joining in vitro are sulphonated to derive the mature functional insulin.
