Define Two-nation Theory. Why did Jinnah who was at first against this,later started supporting this theory?

Jinnah supported this theory because he wanted an independent separate nation for Muslims. This new nation would safeguard and promote Muslim interest. This theory became the basis for the creation of Pakistan.
