Prokaryotes lack membrane bound organelles including the nucleus, chloroplast, mitochondria etc., unlike that of the prokaryotes. The chromosome of a prokaryote is usually circular lacking centromeres and introns. While the eukaryotic chromosome is linear with centromere and kinetochores. They have exons intervened by introns.
Thus both differ vastly in their basic cellular mechanisms especially in transcription and translation.
