The reason dNTPs are used in the Sanger’s method is to terminate the replication process. Every time a dNTP is inserted into the DNA sequence the replication process gets terminated. This generates random sized DNA fragments. After several rounds of these fragments can be analyzed and used to determine the sequence of the DNA.
