Explain an activity to observe a prepared temporary mount of onion peel.

Explanation :

  1. A small sample of tissue can be taken from the onion peel to make a slide and stained with a suitable dye such as safranin and be viewed under the microscope.
  2. Glycerine has a high refractive index and thus can be used in the slide for better vision.

Materials required:

  1. Forceps
  2. Needle
  3. Onion
  4. Slide
  5. Coverslip
  6. Glycerin
  7. Safranin
  8. Microscope

Experiment:

  1. Peel off a thin peel from the onion bulb with the help of forceps and a needle.
  2. Place the peel on a slide.
  3. Put a drop of glycerin on it and also put a drop of orange safranin solution on it.
  4. Cover it carefully with the help of a needle with a coverslip to avoid any air bubbles.
  5. Observe it under a microscope of appropriate power.
