Explain the preparation of copper sulphaate solution that is required for protien test.

Test for protein with coppersulphate is called biuret test.
The required reagents are
  • Sodium Hydroxide and Copper Sulphate Solutions
The steps involved are
  1. Take a small quantity of the food items to be tested.
  2. Grind/mash/paste/powder the food item.
  3. Put the food item into a test tube.
  4. Add the copper sulphate reagent.
  5. Observe the colour of the mixture.
  6. Violet colour indicates the presence of proteins in the tested food item

The method of preparation of copper sulphate reagent is
  1. Take a clean test tube and add 10 ml water.
  2. Using a dropper add two drops of copper sulphate solution to the test tube. If coppersulphate solution is not available, then add solid CuSO4 (copper sulphate) into some water and then make a saturated solution.
  3. Add 10 drops of caustic soda(sodium hydroxide) solution and shake the test tube.
  4. You have a coppersulphate reagent.
