A genomic library is prepared from the total DNA of a cell
line or tissue, whereas acDNA library represents the population of
mRNAs in a tissue.
Genomic DNA libraries are
prepared by performing partial digestion of total DNA with a restriction
enzyme that cuts DNA frequently. On the other hand, cDNA libraries are
prepared by first isolating all the mRNAs in a tissue and then copying
these molecules into double-stranded DNA, using the enzymes reverse
transcriptase and DNA polymerase.
