Method of separation of leaf pigments-: Paper Chromatography is used to separate leaf pigments. It is based on the different rates of adsorption on a suitable adsorbent.

Steps involved in chromatography-

  1. The mixture (leaf pigment) is first dissolved in a substance called the mobile phase and then it is spotted on the chromatogram paper (stationary phase) and dried.
  2. The paper is then placed in a container having a solvent such that only the tip of the paper touches the solvent and not the bottom of the container. The solvent is adsorbed by the paper and due to capillary action, the solvent moves up.
  3. As the pigments dissolve in and move with the solvent, the solvent also carries plant pigment extract on the chromatogram.
  4. The leaf pigments separate themselves on the basis of their differences in their affinity towards stationary and mobile phases.
