The laboratory setup comprises of ammonium chloride which is heated with slaked lime in a test tube:
2NH4Cl(s)+Ca(OH)2(s)CaCl2(s)+2H2O(g)+2NH3(g)

As the reactants are heated, ammonia along with water vapours and calcium chloride is formed.
This mixture is then passed through quicklime which acts as a drying agent and absorbs the water vapours formed in the above reaction.
Being lighter than air, ammonia gas is then collected in an inverted cylindrical column by downward displacement of air.
Also, ammonia gas is basic in nature which can turn a moist red litmus paper blue.
