DNA unwinding by helicases creates topological stress/supercoils in the helical DNA structure which is relieved by the action of topoisomerases. DNA gyrase is topoisomerase II that can introduce negative supercoils using the energy of ATP. It cleaves both strands of a DNA molecule and passes another duplex through the break. Topoisomerases and gyrase are present in the nucleus and are associated with ribosome which makes option A and B wrong. The peptidyl transferase forms peptide bond between the carboxyl group of peptidyl-tRNA present at P site and the free amino group of peptidyl-tRNA present at A site. As we know that ribosomes are the site of protein synthesis and have two subunits; large and small. The peptidyl transferase activity is the integral factor associated with large ribosome subunit. The correct option is C. RNA polymerase enzyme adds ribonucleotides to synthesize RNA using the base sequence of template DNA strand which makes option D incorrect.
