Question

QUESTION: Length of double stranded DNA is 340 Å . It contains 20% Adenine . Calculate total no of hydrogen bonds. QUERY: It is the question and I am confused what approach to take! If I do it by converting it into nucleotide I am getting 260 hydrogen bonds and if by taking that 20 basepairs out of 100 are adenine thymine base pairs and that then the answer comes to be different! What approach should I take?

Solution

Distance between consecutive bases is 3.4 Å

340 Å means it has 340/3.4 = 100 base pairs ( that means 100x2 = 200 bases)

Since it has 20% cytosine it will also have 20% guanine ( cytosine always form bond with guanine).

So rest 60% is adenine and thymidine and they are complementary they both are 30% each.

So 20% of 200 is 40 .

No. Of cytosine bases is 40 each forming 3 H-bonds hence 120 H-bonds.

No. Of adenine bases is 60 each forming 2 H-bonds hence 120 H-bonds.

Total H-bonds is 120+120 = 240.
