An antibiotic resistance gene is a selectable marker. A selectable marker is a gene present in vectors such as plasmids that help in identifying and eliminating non transformants and selectively permitting the growth of the transformants. During such selection using antibiotic resistance genes, antibiotics kill bacteria that do not have the plasmid vector with the resistance gene in the cell, hence selecting the transformants.

DNA ligase is used to link the antibiotic resistance gene with the plasmid. DNA ligase catalyses the joining of the 3′-OH to the 5′-phosphate, by forming a phosphodiester bond between two successive nucleotides.
