When a living tissue (plant tissue or animal tissue) is ground in a mortar and pestle along with trichloroacetic acid (TCA), the lipids form vesicles which are insoluble in acid.

These vesicles are insoluble in water as well. So they don’t dissolve in acid and hence do not flow down along with the filtrate (acid soluble pool) and get retained in the retentate (acid insoluble pool).

The molecular weight (MW) of lipids is found to be <800 Da.
