The girdling experiment proves that ascent of sap occurs through the xylem elements, which are embedded deep inside the tree trunk, whereas transport of sugars occurs through the secondary phloem, which is present in the bark of the tree.
Cobalt-chloride paper test is the hygrometric method to demonstrate transpiration. Change in colour of the filter paper slip from blue to pink indicates transpiration. As considerably more stomata are present on the lower side of the leaf, more transpiration will occur on the lower surface of the leaf.
