During the process of glycolysis, substrate level phosphorylation of ADP as well as dephosphorylation of ATP occurs. There is dephosphorylation of 2 ATP to ADP and phosphorylation of 4 ADP to ATP, so there is a net gain of 2 ATP in glycolysis.
