Nucleolus was discovered by Fontana in 1781 and given the present name by Bowman, 1840. It contains four parts- (a) Amorphous (b) Fibrillar (c) Granular (d) Chromatin.
Both fibrillar and granular part are made up of RNA and protein. The nucleolus is the site of rRNA synthesis, which occurs on DNA template of certain chromosomes. Newer studies exclude DNA from the nucleolus and consider the nucleolus as a rounded body that is usually basophilic and eccentrically placed in the nucleus. Although originally classified under chromatin because of its basophilic nature, it was later excluded from the most strict definition of the term by its failure to stain with the Feulgen reaction for DNA, and by the fact that it was shown instead to be rich in RNA.
