-Iodine test is to detect the presence of starch. Chloroplasts are responsible to produce it by photosynthesis. If chloroplasts are removed, no photosynthesis takes place and hence, no starch will be formed. The leaves will answer a negative test for starch.
-The cell membrane also known as the plasma membrane is a selectively permeable membrane. It allows only selected substances (solids and liquids) to pass through it. If the cell membrane is removed, there will be no check on the substances entering and exiting the cell.
