In sickle-cell anaemia, the RBCs become sickle-shaped. It is caused due to point mutation (substitution) in the gene that codes for beta haemoglobin chain. Due to this mutation glutamic acid is replaced by valine at the 6th position of the beta haemoglobin chain.
