In recombinant DNA technology, in order to extract DNA from the cell, the cell is treated with various enzymes to dissolve the cell wall (if present), cell membrane and digest the macromolecules (proteins) associated with the DNA.

The purified DNA can be seen as a collection of fine threads which can be scooped and swirled around a glass rod or wooden spooling stick. This process is called spooling. This helps to isolate the purified DNA threads from solutions.
