Spooling is the process of ______.
collecting isolated DNA
DNA spooling is a method of obtaining DNA in a form of spool over a glass rod. This forms the last step of DNA extraction. During isolation of DNA, ethanol is added in the last step to precipitate DNA, after that the DNA can be spooled on a glass rod. This is achieved by inserting a thin glass rod in the tube having precipitated DNA; the glass rod is gently rotated with the help of fingers and DNA adheres to the glass rod.
Polymerase chain reaction is the process of amplifying a section of DNA which normally contains genes of interest.
Gel purification is a standard procedure to recover the separated DNA molecule from agarose after electrophoresis. Addition of salt buffer most likely disrupts the cation bridge, thus DNA is eluted or freed from the gel.
Fragments of DNA separated after gel electrophoresis are transferred to nylon or nitrocellulose membrane in a process termed as Southern blotting. The blotting process helps to detect specific DNA sequences in the samples.
