Totipotency is the ability of a cell to divide. F.C steward was first to illustrate the capacity of cellular totipotency. He stated that cells derived from the plant parts like root, stem, leaf, vegetative bud, floral bud, anther and embryo can exhibit cellular totipotency when cultured under the controlled aseptic condition on a nutrient media. He took slices of phloem tissue of carrot and cultured it on nutrient media. It turned to callus. Callus when treated with auxin it promotes rooting and when it is treated with cytokinin it stimulates cell maturation, differentiation and finally, promotes the development of stem. This experiment proved that cells of the plant retain the capacity to divide.
