Danish bacteriologist Christian Gram, for the first time, classified bacteria on the basis of their cell walls into two groups: Gram positive and Gram negative, by staining with crystal violet and safranin.
The cell wall of Gram positive bacteria is less complex with peptidoglycan compounds and proteins and no lipids, whereas in Gram negative bacteria, cell walls are more complex with peptidoglycan compounds, phospholipids and lipopolysaccharides and contain 20% lipids.
