Totipotency of a cell is its ability to divide and differentiate into any cell type, such that it can develop into an entire organism.

In a mature plant, meristematic tissues actively divide and are essentially totipotent cells. But permanent tissues are those who have lost the ability to divide and have differentiated into a specific cell type.
In order to induce totipotency the permanent tissues must lose their function and start dividing (dedifferentiation).

Tissue culture is the technique of growing plant cells, tissues or organs in a sterile culture medium under controlled environmental conditions. In this technique certain plant parts, cell or tissue (explant) can bemade to develop into a whole plant by culturing it in a medium containing the appropriate nutrients and the right concentration of growth hormones.
