The Southern hybridisation/ blotting technique was devised by E. M. Southern in 1975. In this technique, the DNA molecules are transferred and immobilised on a nitrocellulose or nylon membrane from an electrophoresis gel in such a way that the pattern present in the gel can be reproduced on the membrane. This is followed by treatment with labelled probes which end up binding to its complementary sequence in the sample.
