  2. The principle of ELISA is antigen-antibody interaction.
  3. There are four types of ELISA namely Direct ELISA (antigen-coated plate; screening antibody), Indirect ELISA (antigen-coated plate; screening antigen/antibody), Sandwich ELISA (antibody-coated plate; screening antigen), and Competitive ELISA (screening antibody)

The sensitivity of the Sandwich ELISA depends on four factors:

  1. Avidity of the first antibody for the antigen.
  2. Solid-phase bound with the first antibody molecules number
  3. Avidity of the second antibody for the antigen.
  4. The particular activity of the second antibody.
