Due to changes in pH, temperature, presence of certain salts/chemical agents, the complex-three dimensional structure (native conformation) of protein is disrupted. Due to this, protein loses its biological activity. This is called denaturation of protein.
Two diseases caused due to deficiency of protein are kwashiorkor and marasmus.
