Plasmid:

  1. Plasmid is the circular, extrachromosomal foreign DNA which have the ability to replicate independently.
  2. They have antibiotic resistant genes present in them.

Plasmid amplification:

  1. Amplification is used for enhancing the copy number of a plasmid.
  2. Some plasmids have capability to replicate even in the absence of protein synthesis.
  3. This is incompatible with the primary bacterial chromosome, which are not able to replicate in the given environment.
  4. This characteristic can be used during the multiplication of bacterial culture for plasmid DNA purification.
  5. As a result, plasmid amplification is a particularly effective method for increasing the production of multicopy plasmids.
