Ligase is used for joining purpose of recombinant DNA:ligaseis an enzyme thatcan catalyse thejoining of two large molecules by forming a new chemical bond, usually with accompanying hydrolysisof a small pendant chemical group on one of the larger molecules or the enzyme catalyzing the linking together of two compounds, e.g., enzymes that catalyze joining of C-O, C-S, C-N, etc. In general, a ligase catalyzes the following reaction:

Ab + C → A–C + b

or sometimes

Ab + cD → A–D + b + c + d + e + f

where the lowercase letters signify the small, dependent groups. Ligase can join two complementary fragments of nucleic acid and repair single stranded breaks that arise in double stranded DNA during replication.

polymeraseis an enzyme(EC 2.7.7.6/7/19/48/49) that synthesizes long chains or polymersof nucleic acid. DNA polymeraseand RNA polymerase areused to assemble DNAand RNAmolecules, respectively, by copying a DNA or RNA template strand using base:pairinginteractions.

A polymerase from the thermophilic bacterium(Taq) , EC 2.7.7.7) is used in the polymerase chain reactionan important technique of molecular biology
