The Frederick Sanger method of DNA sequencing using automated DNA sequencers is based on the principle of selective incorporation of the chain-elongating inhibitors of DNA polymerase (chain-terminating dideoxynucleotides) via enzyme DNA polymerase during in vitro replication of DNA.
