If antibiotic is not added to the medium in which a recombinant is growing it won’t be possible to select the recombinant bacteria from a mix of recombinant and non-recombinant bacteria. If we grow the bacteria on a solid media which contains an antibiotic, only the cells that were transformed with a plasmid of our choice containing the gene for antibiotic resistance will grow and the others will die making it possible for us to screen the recombinant bacteria.
