NADP reductase enzyme is located on the outer side of the thylakoid membrane of the chloroplast.

Role of NADP reductase in a proton gradient

NADP reductase reduces NADP to NADPH+H+.
● The protons released by the reduction of NADP are pumped inside the thylakoid lumen.
● As a result, the concentration of protons increases in the thylakoid lumen as compared to the outside of the thylakoid.
● This leads to the formation of the proton gradient.
