Which group of elements was missing from Mendeleev's original periodic table ?

Group (or family) 8; the noble gases. They are naturally very unreactive to almost any chemical or element, although their are some exceptions (like fluorine). The best way to find or identify a chemical is to react it with something else, but because they are so unreactive, they weren't found for a while.
