Polymerase chain reaction:

  1. In molecular biology, the PCR, or polymerase chain reaction, is a method used to produce many copies of a specific DNA region.
  2. These copies allow scientists to study that small part of DNA in detail.
  3. The process starts with Denaturation.
  4. The second step in polymerase is annealing or hybridization.
  5. Lastly, the elongation process is followed.

Annealing:

  1. Tag polymerase requires a short RNA fragment to initiate replication of the DNA synthesized by RNA polymerase in a typical cell.
  2. A complementary double-stranded oligo with a short PCR reaction is added.
  3. It binds to denatured DNA and serves as the starting point for the replication process.
  4. Primers (double-stranded oligos) complement the upstream and downstream sequences of the gene of interest.
  5. Two primers are used for each DNA strand.
