In recombinant DNA technology, a gene of interest from one organism is inserted in a different organism with the help of different tools and techniques.

The gene of interest is isolated and ligated with the plasmid first, which is a vector to carry the gene into the host. This rDNA is inserted into the host cell.

Now there are three cases:
  • Host cells that took up neither the plasmid without the insert nor the plasmid with the insert (non-transformants)
  • Host cells that took up just the plasmids without the insert (transformants but non-recombinants)
  • Host cells that took up the rDNA (plamid with the gene insert) (transformants and recombinants)
The plasmid has antibiotic resistance genes on it which are selectable markers for distinguishing between the transformants and non-transformants.

The transformants will grow on antibiotic-containing medium as they have antibiotic resistance genes in them. Whereas, the non-transformants will not survive in the antibiotic-containing medium.

Recognition sites are only used for inserting the gene of interest and ori sites initiate the process of replication in plasmids.
