Due to the permeability of plasma membrane of RBC to anions, HCO−3 diffuse into the blood plasma from RBC at the tissues. The H+ ions are buffered by hemoglobin which turns into HHb(acid hemoglobin).
Hemoglobin acts as a buffer at physiological pH because of its high content of histidine.
As a result, the molecules lose their affinity for oxygen and so O2 is released. It diffuses into the plasma and from there into the tissues.
To maintain electrical balance Cl− ions diffuse from plasma into RBC. This exchange of chloride and bicarbonate ions between RBC and plasma at tissue level is called chloride shift or hamburger's phenomenon.
