The acid-soluble pool consists of substances whose molecular weight is less than 1000 daltons. The substances commonly found in the acid-insoluble pool are called biomicromolecules such as amino acids, monosaccharides and nucleotides. It roughly represents the cytoplasmic content of the cell.
Even though lipids weigh less than 1000 daltons, it is not found in the acid-soluble pool. This is because upon crushing the living tissue, the lipids form vesicles which cannot be dissolved in the acid.
