A cloning vector is a small piece of DNA, taken from a virus, a plasmid, or the cell of a higher organism, that can be stably maintained in an organism, and into which a foreign DNA fragment can be inserted for cloning purposes. An expression plasmid is a circular piece of DNA that contains an origin of replication, the appropriate regulatory regions located 5 to the site in which the gene of interest is inserted, and a criterion for selection. The origin of replication is important to assure the replication of the plasmid within the host bacterium. The pUC18 plasmid and pUC19 plasmid enable successful cloning of larger DNA fragments than the M13mp18 RF Phage Vector. Because these cloning vectors contain a multiple cloning site at the lacZ' region, recombinant plasmids can be verified via blue/white colony screening using agar plates containing IPTG and X-Gal.
