The enzymes that cleave the sugar-phosphate backbone of DNA are called restriction endonucleases. There are three types of restriction endonucleases, namely type I, II, and III. The first isolated restriction endonucleases were Type II restriction endonucleases. These were isolated by Hamilton Smith and Daniel Nathan. They showed that Type II endonucleases are simple, independent of ATP and cleave the DNA within the recognition sequence itself.
