M. Nirenberg, Khorana and Holley were awarded Nobel Prize for in vitro synthesis of polypeptides using artificially synthesized mRNA. Marshall Nirenberg and Heinrich Matthaei incubated synthetic polyuridylate, poly(U), GTP, ATP, E. coli extract and a mixture of the 20 amino acids in 20 different tubes containing a different radioactively labelled amino acid to produce the poly(U) mRNAs. It was concluded that the triplet codon UUU encodes phenylalanine. Khorana used the same approach to reveal that polycytidylate, encodes a polypeptide containing only proline and polyadenylate encodes polylysine. Replica plating technique rapidly transfers microorganism colonies to numerous petri plates and is used for detection of auxotrophs. It was given by Joshua Lederberg & Esther Lederberg; a pattern of colonies growing on the complete medium is transferred to the second plate of the medium that lacks a particular nutrient, for example, methionine. The areas on the second plate where colonies fail to grow confirm that the original colony was a methionine-requiring auxotroph. The correct answer is C.
